PURPOSE: B-5 fixative is routinely used for bone marrow biopsies, and on some<\/span>
\nlymph nodes when lymphomas are suspected.<\/span><\/p>\n

REAGENTS:<\/strong>
\nStock B-5 Solution:
\nMercuric chloride 12.0 gm
\nSodium acetate 2.5 gm
\nDistilled water 200.0 ml
\nMix well, do not use any metal
\nutensils or foil lined lids,
\nsolution is stable for 1 year.
\nInitial and date.
\nWorking Solution:
\nStock solution 20.0 ml
\nFormaldehyde 2.0 ml
\nMix immediately before use.
\nDiscard solution according to the
\nhazardous waste regulations.
\nCAUTION: CARCINOGENIC, IRRITANT,
\nCORROSIVE
\nSAFETY:<\/strong> Work in well ventilated area, wear gloves, lab coat, and goggles.
\nMercuric chloride; severe skin and eye irritant. Target organ effects on
\nreproductive, urogenital, respiratory, gastrointestinal and fetal systems via
\ningestion and inhalation routes. Corrosive to metals.
\nFormaldehyde: severe eye and skin irritant. Sensitizer by skin and respiratory
\ncontact. Toxic by ingestion and inhalation. Target organ effects on
\nrespiratory system. Corrosive. Carcinogen.
\nDISPOSAL: Pour used B-5 fixative in a glass bottle labeled as “B-5 Waste”, do
\nnot pour down the drain. Follow instructions for WASTE REDUCTION in the
\nHistology SOP Manual. To be disposed of by Safety Services.
\nFIXATION TIME: 2 to 4 hours, note time on container, do not leave overnight.
\nPROCEDURE:<\/strong>
\n1. Retain fixed specimen in 10% formalin or 70% alcohol.
\n2. Tissue sections that are B-5 fixed contain mercury deposits and must be
\ntreated to remove the pigments prior to staining, after deparaffinizing:
\na) Lugol\u2019s iodine: 5 minutes, wash.
\nb) 5% hypo: 5 minutes, wash and stain as desired.
