{"id":298529,"date":"2019-03-27T09:56:06","date_gmt":"2019-03-27T08:56:06","guid":{"rendered":"https:\/\/www.deltamicroscopies.com\/produit\/bouins-fixative\/"},"modified":"2025-08-10T19:44:51","modified_gmt":"2025-08-10T18:44:51","slug":"bouins-fixative","status":"publish","type":"product","link":"https:\/\/www.deltamicroscopies.com\/en\/products\/bouins-fixative\/","title":{"rendered":"Bouin&#8217;s Fixative"},"content":{"rendered":"<p><b>Bouin solution<\/b>, or\u00a0<b>Bouin&#8217;s solution<\/b>, is a compound\u00a0fixative\u00a0used in\u00a0histology.<sup id=\"cite_ref-1\" class=\"reference\"><\/sup>\u00a0It was invented by French biologist\u00a0<a class=\"new\" title=\"Pol Bouin (page does not exist)\" href=\"https:\/\/en.wikipedia.org\/w\/index.php?title=Pol_Bouin&amp;action=edit&amp;redlink=1\">Pol Bouin<\/a>\u00a0and is composed of\u00a0picric acid,\u00a0acetic acid\u00a0and\u00a0formaldehyde\u00a0in an aqueous solution.\u00a0<sup id=\"cite_ref-2\" class=\"reference\"><\/sup>\u00a0Bouin&#8217;s fluid is especially useful for fixation of\u00a0gastrointestinal tract\u00a0biopsies because this fixative allows crisper and better\u00a0nuclear\u00a0staining\u00a0than 10% neutral-buffered formalin. It is not a good fixative when tissue\u00a0ultrastructure\u00a0must be preserved for\u00a0electron microscopy. However, it is a good fixative when tissue structure with a soft and delicate texture must be preserved. The acetic acid in this fixative\u00a0<a title=\"Lysis\" href=\"https:\/\/en.wikipedia.org\/wiki\/Lysis\">l<\/a>yses\u00a0red blood cells and dissolves small iron and calcium deposits in tissue.<\/p>\n<h2><span id=\"Variations\" class=\"mw-headline\" style=\"font-size: 14pt;\">Variations Bouin<\/span><\/h2>\n<h3><span id=\"Gendre_solution\" class=\"mw-headline\" style=\"font-size: 12pt;\">Gendre solution<\/span><\/h3>\n<p><span style=\"font-size: 12pt;\">Gendre solution is an alcoholic versi<\/span>on of Bouin solution. An alcoholic solution saturated with picric acid is used instead of an aqueous solution saturated with picric acid when making this solution. This solution is useful when glycogen and other carbohydrates must be preserved in tissue. It is prepared by a mixing saturated solution of picric acid in 95% ethanol (80ml) with formalin (37-40% formaldehyde) (15 ml) and glacial acetic acid (5ml).\u00a0<sup id=\"cite_ref-8\" class=\"reference\"><\/sup>\u00a0Gendre&#8217;s fixative contains more picric acid than Bouin&#8217;s, because of greater solubility (6.23%w\/v) in ethanol than in water (1.23%w\/v).<sup id=\"cite_ref-9\" class=\"reference\"><\/sup><\/p>\n<h3><span id=\"Hollande_solution\" class=\"mw-headline\" style=\"font-size: 12pt;\">Hollande solution<\/span><\/h3>\n<p>Hollande solution is a version of Bouin solution that contains\u00a0copper acetate.
