The addition of\u00a0Latex Spheres to the Grating Replica\u00a0provides a double check of the accuracy of magnification calibration. It is particularly useful at higher magnifications where only a few squares of the replica pattern can be photographed together.<\/p>\n

Note:<\/strong>\u00a0The latex particles in this specimen have a diameter of 0.261 micron.<\/p>\n

To use both the Replica and Latex Spheres for Magnification Calibration:<\/h4>\n\n\n\n\n\n\n
(1)\u00a0 Calculate the magnification using the diffraction grating replica pattern by the\u00a0 following formula:<\/td>\n<\/tr>\n
Magnification<\/td>\n=<\/td>\nDistance in mm between limiting lines x 2,160 \/
\nNumber of spaces between limiting lines<\/td>\n<\/tr>\n
(2)\u00a0 Calculate the magnification by measuring the magnified image of the 0.262 micron diameter latex spheres,\u00a0 applying the following formula:<\/td>\n<\/tr>\n
Magnification<\/td>\n=<\/td>\nDiameter (mm) x 3831.4<\/td>\n<\/tr>\n<\/tbody>\n<\/table>\n

(Measure as many latex spheres as possible and calculate the average diameter. Ignore any latex spheres which are distorted.)<\/p>\n

Care of Grating Replica Specimen:<\/h4>\n

When not in use the replica should be kept in a dust-free atmosphere (as the same vial in which it is supplied).\u00a0\u00a0NEVER TRY TO CLEAN IT<\/strong>.\u00a0 Avoid bending or touching thereplica.\u00a0 When viewing a replica specimen in the TEM always begin at low magnification, with one square of the supporting copper grid filling the EM screen, slowly increase the illumination to near maximum intensity (but not to cross-over) then reduce the level of the illumination and go to the desired magnification.\u00a0 Repeat this procedure every time a new area of the replica is to be viewed.<\/p>\n","protected":false},"excerpt":{"rendered":"
