Important parameters for your sample preparation

Good sample preparation for microscopic analysis is necessary to keep tissue components as close to life as possible. For optimum conservation of biological samples, the sampling conditions are as important as the fixation itself.

Different factors influence the selection and fixation efficiency of a fixative, including sample size, osmolarity, pH, penetration rates of reagents such as fixer and sample depth or thickness , the fixing temperature, the fixing time, the concentration of the fixatives, the fixing steps.

As a result, several factors come into play when choosing and adapting a protocol based on the sample being processed. A cautious Microscopist will say that there are as many protocols or procedures for preparation as samples. A universal method exists but it has to be adapted to the treated sample:

hardness, porosity, osmolarity …
choice of solvents for dehydration and impregnation with resins in order to avoid extractions of the elements of the sample.
choice of resins: epoxy, acrylic, ….
